Biological Background: FFAR4 Function and Localization
- FFAR4 (Free fatty acid receptor 4), also known as GPR120, GPR129, O3FAR1, and by other aliases, is a G protein-coupled receptor for long-chain fatty acids.
- It signals via phosphoinositidase C-linked G proteins GNAQ/GNA11 (Gq/G11) and beta-arrestin pathways, modulating intracellular calcium, cAMP, and MAP kinases. Related references: PMID:22282525, PMID:22343897, PMID:24742677
- Upon ligand binding, FFAR4 associates with beta-arrestin ARRB2, which couples the receptor to specific downstream signaling and mediates receptor endocytosis. Related references: PMID:22282525, PMID:24817122
- FFAR4 mediates the anti-inflammatory effects of omega-3 PUFAs by inhibiting NLRP3 inflammasome activation through ARRB2 association. Related references: PMID:23809162

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 333–377 of human FFAR4, a C-terminal region. This design may enable detection of multiple isoforms.
- For western blotting, a band around 40 kDa is expected. Validate using known positive tissues such as adipose tissue or pancreatic islet cell lines.
- In ELISA, consider using purified recombinant FFAR4 protein as a standard to optimize antibody concentration.
- Cross-reactivity with mouse and rat FFAR4 is indicated; verify in the relevant species model as needed.
