Biological Background: FFAR3 Function and Localization
- FFAR3, also known as G protein-coupled receptor 41 (GPR41), is a free fatty acid receptor 3 that belongs to the G protein-coupled receptor family. It is activated by short-chain fatty acids (SCFAs) such as propionate, butyrate, and pentanoate, which are major products of dietary fiber digestion by gut microbiota. Related references: PMID:12496283, PMID:12711604
- The receptor couples to pertussis toxin-sensitive G(i/o)-alpha proteins, leading to inositol 1,4,5-trisphosphate formation, intracellular calcium mobilization, MAPK1/ERK2 and MAPK3/ERK1 phosphorylation, and inhibition of intracellular cAMP accumulation. Related references: PMID:12711604
- FFAR3 plays a role in whole-body energy homeostasis by regulating sympathetic neuron activity, energy expenditure, heart rate, blood pressure via renin secretion, and hormone production (e.g., PYY peptide and leptin), as well as intestinal gluconeogenesis and glucose homeostasis.
- In the gut, FFAR3 contributes to intestinal immunity by mediating SCFA-induced rapid production of chemokines and cytokines by intestinal epithelial cells, thus regulating inflammatory and immune responses.
- Subcellular localization: The receptor is a multi-pass membrane protein found on the cell membrane (plasma membrane).
- Tissue specificity: Highest expression in adipose tissue, with lower expression across all tissues. It is also expressed in sympathetic ganglia, consistent with its role in sympathetic neuron modulation.
- Keywords include: 3D-structure, Cell membrane, Disulfide bond, G protein-coupled receptor, Glycoprotein, Immunity, Inflammatory response, Lipid-binding, Membrane, Receptor, Reference proteome, Transducer, Transmembrane, Transmembrane helix. Molecular weight: 38649 Da (calculated from sequence).
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal region (aa 257-346) of human FFAR3, which is expected to recognize both the human protein and cross-react with mouse and rat orthologs, as indicated. However, for the best results, validate the antibody in the specific sample system.
- For Western blot (WB) analysis, consider using whole-cell lysates from tissues with high FFAR3 expression, such as adipose tissue or sympathetic ganglia. The predicted molecular weight is approximately 39 kDa; ensure appropriate gel percentage and transfer conditions for a multi-pass transmembrane protein.
- For ELISA applications, the antibody’s polyclonal nature provides robust detection. Consider testing different coating antigen concentrations and detection methods to optimize signal-to-noise ratio.
- Since FFAR3 is a GPCR with seven transmembrane domains, sample preparation (e.g., using mild detergents, avoiding boiling) may be critical to preserve epitope recognition.
