Biological Background: Ferritin Light Chain Function and Localization

  • Ferritin light chain (FTL) is the light subunit of ferritin, the primary iron storage protein in cells (UniProt P02792).
  • It sequesters iron in a non-toxic, bioavailable form by storing ferric hydroxides after oxidizing ferrous iron, thereby maintaining iron homeostasis.
  • FTL facilitates iron delivery to cells, including mediating uptake in developing kidney capsule cells, as indicated by similarity studies.
  • Ferritin light chain transports iron to lysosomes via interaction with the cargo receptor NCOA4, where autophagic degradation releases iron for cellular utilization. Related references: PMID:24695223
  • Subcellularly, FTL localizes to the cytoplasm, cytoplasmic vesicles, autophagosomes, and autolysosomes, reflecting its dynamic role in iron trafficking and degradation.
