Biological Background: Ferritin Light Chain Function and Localization
- Ferritin light chain (FTL) is the light subunit of ferritin, the primary iron storage protein in cells (UniProt P02792).
- It sequesters iron in a non-toxic, bioavailable form by storing ferric hydroxides after oxidizing ferrous iron, thereby maintaining iron homeostasis.
- FTL facilitates iron delivery to cells, including mediating uptake in developing kidney capsule cells, as indicated by similarity studies.
- Ferritin light chain transports iron to lysosomes via interaction with the cargo receptor NCOA4, where autophagic degradation releases iron for cellular utilization. Related references: PMID:24695223
- Subcellularly, FTL localizes to the cytoplasm, cytoplasmic vesicles, autophagosomes, and autolysosomes, reflecting its dynamic role in iron trafficking and degradation.
- Mutations in FTL are associated with neurodegenerative disorder neuroferritinopathy (NBIA3) and L-ferritin deficiency (LFTD), as well as cataract formation.

- The immunogen covers amino acids 1–175, corresponding to the full-length mature protein; thus, the antibody may recognize both native and conformationally altered forms in denaturing and non-denaturing assays.
- For Western blot, a band around 20 kDa is expected in lysates from human, mouse, or rat tissues/cells; include a loading control and verify with a known positive sample.
- In immunohistochemistry (paraffin), antigen retrieval may enhance epitope accessibility; titrate antibody and include appropriate negative controls to confirm specificity.
- For ELISA, determine the optimal working concentration by checkerboard titration against the recombinant immunogen or target protein.
