Biological Background: Ferritin Heavy Chain Function and Localization
- Ferritin heavy chain (FTH1, also known as FTH, FTHL6, cell proliferation-inducing gene 15 protein) is a key iron storage protein belonging to the ferritin family. It assembles into a 24-subunit shell to store iron in a soluble, non-toxic form.
- The protein possesses ferroxidase activity, catalyzing the oxidation of Fe²⁺ to Fe³⁺ and thereby facilitating iron deposition as ferric hydroxides within the ferritin cage (PubMed:9003196).
Related references: PMID:9003196 - Beyond storage, ferritin heavy chain mediates iron delivery to cells, including specialized uptake in developing kidney capsule cells, and participates in intracellular iron trafficking.
- Its lysosomal degradation is regulated by the cargo receptor NCOA4, which directs ferritin to autophagosomes for iron release—a process critical for cellular iron homeostasis (PubMed:24695223, PubMed:26436293).
Related references: PMID:24695223 PMID:26436293 - Subcellular localization includes the cytoplasm, lysosomes, and cytoplasmic vesicles such as autophagosomes, reflecting its dynamic role in iron storage and recycling.
- The protein is expressed in the liver, a major iron storage organ, and its expression can be regulated by iron levels and oxidative stress.
- Post-translational modifications including acetylation and phosphorylation, along with its oxidoreductase activity, fine-tune ferritin function and stability; the molecule is also implicated in neurodegeneration.
Experimental Guidance and Technical Tips
- The immunogen comprises the full-length human ferritin heavy chain (aa 1–183), making this polyclonal antibody suitable for detecting native protein conformations across multiple applications.
- For Western blotting, the predicted molecular weight is approximately 21 kDa; consider using a 12–15% SDS-PAGE gel for optimal separation.
- In immunohistochemistry (IHC-P), antigen retrieval conditions (e.g., citrate buffer pH 6.0 or EDTA pH 9.0) should be optimized for each tissue type; validate with appropriate positive and negative controls.
- When using immunofluorescence (IF/ICC), fixation and permeabilization methods may influence signal; test methanol or paraformaldehyde fixation with Triton X-100 permeabilization.
- For ELISA, the antibody can be used as a capture or detection reagent; pairing with a monoclonal antibody against a non-overlapping epitope may improve assay performance—validate in the relevant sample system.
