Biological Background: Ferritin Heavy Chain Function and Localization
- Ferritin heavy chain stores iron in a soluble, non-toxic form and exhibits ferroxidase activity, oxidizing ferrous iron to ferric hydroxides for safe deposition. Related references: PMID:9003196
- It mediates iron delivery to cells and plays a role in iron uptake in capsule cells of the developing kidney (By similarity).
- Iron release is regulated via NCOA4-mediated delivery to lysosomes for autophagic degradation. Related references: PMID:24695223 PMID:26436293
- Primarily localized in the cytoplasm, but also found in lysosomes, autophagosomes, and cytoplasmic vesicles.
- Post-translational modifications include acetylation and phosphorylation; linked to neurodegeneration.
- Expressed in the liver; part of the ferritin family with a predicted molecular weight of ~21 kDa.
Experimental Guidance and Technical Tips
- The antibody was raised against a synthetic peptide derived from human Ferritin Heavy Chain. Cross-reactivity with mouse and rat suggests conserved epitope recognition; consider sequence alignment to verify.
- For Western blotting, the target protein runs at ~21 kDa. Use liver lysates as a positive control given its expression profile.
- In immunofluorescence and immunohistochemistry, permeabilization steps may be required due to the cytoplasmic localization of the protein.
- For ELISA applications, consider pairing with a detection antibody against a distant epitope for sandwich assay development. Validate in the relevant sample matrix.
