Biological Background: Ferredoxin Reductase Function and Localization
- Ferredoxin Reductase, officially NADPH:adrenodoxin oxidoreductase (mitochondrial), is encoded by the FDXR gene and is also known as ferredoxin--NADP(+) reductase.
- Serves as the first electron transfer protein in all mitochondrial P450 systems, including cholesterol side-chain cleavage in steroidogenic tissues, steroid 11-beta hydroxylation in the adrenal cortex, 25-OH-vitamin D3-24 hydroxylation in the kidney, and sterol C-27 hydroxylation in the liver.
- Acts as a ferredoxin--NADP(+) reductase essential for coenzyme Q biosynthesis; together with FDX2, transfers electrons required for the hydroxylation reaction performed by COQ6. Related references: PMID:38425362
- Localizes to the mitochondrion inner membrane, consistent with its role in mitochondrial electron transport.
- Belongs to the flavoprotein family and utilizes FAD and NADP cofactors, functioning as an oxidoreductase in electron transport.
- Involved in lipid and steroid metabolism, with genetic variants associated with diseases like deafness, neurodegeneration, and neuropathy, as highlighted by UniProt keywords.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein spanning amino acids 142–491 of human Ferredoxin Reductase, covering a large portion of the mature protein; consider this when designing epitope mapping or competitive assays.
- For western blot (WB), the predicted molecular weight is approximately 54 kDa; validate in your sample system as mobility may vary due to post-translational modifications or processing.
- In immunofluorescence (IF/ICC and IF-P) and immunohistochemistry (IHC-P), expect mitochondrial staining; include appropriate organelle markers to confirm localization.
- For ELISA applications, the antibody’s performance may depend on the plate coating and detection format; we recommend optimizing concentrations and incubation conditions for your specific assay setup.
