Biological Background: Ferredoxin Reductase Function and Localization
- Ferredoxin reductase (FDXR), also known as adrenodoxin reductase (ADR/ADXR), is a mitochondrial flavoprotein that serves as the first electron transfer protein in all mitochondrial cytochrome P450 systems.
- It plays a critical role in cholesterol side-chain cleavage for steroidogenesis in steroidogenic tissues, steroid 11-beta hydroxylation in the adrenal cortex, vitamin D metabolism in the kidney, and sterol C-27 hydroxylation in the liver (By similarity).
- Together with ferredoxin FDX2, FDXR transfers electrons to COQ6 for the hydroxylation step essential in coenzyme Q biosynthesis.
Related references:
PMID:38425362 - FDXR is localized to the mitochondrial inner membrane, consistent with its role in electron transport chains.
- The protein is associated with several key biological processes, including cholesterol and steroid metabolism, lipid metabolism, and electron transport, as indicated by UniProt keywords.
- Additionally, FDXR has been linked to disease variants causing deafness, neurodegeneration, and neuropathy, highlighting its clinical relevance.
Experimental Guidance and Technical Tips
- For Western blotting, a band at approximately 54 kDa is expected. Consider optimizing blocking and antibody dilution for your specific sample type.
- In immunofluorescence (IF-P), the antibody is expected to stain mitochondria; co-staining with mitochondrial markers can confirm localization.
- The immunogen corresponds to amino acids 142-491, covering a large portion of the protein, which may provide robust detection across species.
- Validate the antibody in the relevant sample system, such as steroidogenic tissues or cell lines, to confirm specificity.
