Biological Background: Fermitin family homolog 2 Function and Localization
- Fermitin family homolog 2 (FERMT2), also known as Kindlin-2, MIG-2, and PLEKHC1, is a scaffolding protein encoded by the FERMT2 gene in humans.
- Core function: FERMT2 enhances integrin activation mediated by TLN1/TLN2, though it activates integrins weakly by itself. It binds to phosphoinositide-rich membranes and is essential for integrin-mediated cell adhesion, cell spreading, and focal adhesion assembly.
- It participates in the connection between extracellular matrix adhesion sites and the actin cytoskeleton, orchestrating actin assembly and cell shape modulation, and recruits FBLIM1 to focal adhesions.
- FERMT2 plays a role in TGFB1 and integrin signaling pathways, stabilizes active CTNNB1, and is involved in the regulation of Wnt signaling transcription mediated by CTNNB1/TCF7L2.
- Subcellular localization: FERMT2 is found in the cytoplasm, at cell cortex, along stress fibers, at focal adhesions, on lamellipodium membrane, in the nucleus, and at the I band of sarcomeres in myofibrils. It also localizes to the cell surface.
- Tissue distribution: Ubiquitously expressed, with notable presence in numerous tumor tissues, indicating a broad functional role.
- As a phosphoprotein and alternative splicing product, FERMT2 undergoes post-translational modifications that may regulate its activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 380–550 of human FERMT2, a region that may contain conserved motifs across species, consistent with the reported cross-reactivity with mouse and rat.
- For Western blotting, a band near the predicted molecular weight of 78 kDa is expected; consider using positive control lysates from human, mouse, or rat cell lines expressing FERMT2.
- In immunohistochemistry (paraffin-embedded sections), validate staining in tissues known to express FERMT2 (e.g., tumor tissues or tissues rich in focal adhesions), and optimize antigen retrieval conditions.
- For ELISA, this polyclonal antibody can be explored as a capture or detection reagent; however, we recommend performing a titration experiment and validating specificity using recombinant FERMT2 protein or verified positive samples.
- As with any polyclonal antibody, lot-to-lot variability may occur; it is advisable to test each new batch in the intended application with appropriate reference controls.
