Biological Background: FEN-1 Function and Localization

  • Encodes Flap endonuclease 1 (FEN-1), also known as DNase IV, Flap structure-specific endonuclease 1, and Maturation factor 1 (MF1).
  • Functions as a structure-specific nuclease with both 5'-flap endonuclease and 5'-3' exonuclease activities, essential for DNA replication and repair.
  • During DNA replication, cleaves the 5'-overhanging flap structure generated by displacement synthesis, allowing ligation of Okazaki fragments.
  • Participates in long patch base excision repair (LP-BER) by cleaving within apurinic/apyrimidinic (AP) site-terminated flaps.
  • Acts as a genome stabilization factor, preventing DNA secondary structures that could lead to duplications and deletions.
  • Also exhibits 5'-3' exonuclease activity on nicked/gapped dsDNA, RNase H activity, and is involved in rDNA and mitochondrial DNA repair.
  • Localizes to the nucleus, nucleolus, nucleoplasm, and mitochondrion.
