- Encodes Flap endonuclease 1 (FEN-1), also known as DNase IV, Flap structure-specific endonuclease 1, and Maturation factor 1 (MF1).
- Functions as a structure-specific nuclease with both 5'-flap endonuclease and 5'-3' exonuclease activities, essential for DNA replication and repair.
- During DNA replication, cleaves the 5'-overhanging flap structure generated by displacement synthesis, allowing ligation of Okazaki fragments.
- Participates in long patch base excision repair (LP-BER) by cleaving within apurinic/apyrimidinic (AP) site-terminated flaps.
- Acts as a genome stabilization factor, preventing DNA secondary structures that could lead to duplications and deletions.
- Also exhibits 5'-3' exonuclease activity on nicked/gapped dsDNA, RNase H activity, and is involved in rDNA and mitochondrial DNA repair.
- Localizes to the nucleus, nucleolus, nucleoplasm, and mitochondrion.
- Subject to acetylation, methylation, and phosphorylation, with a molecular weight of ~43 kDa (calculated 42.6 kDa).
