  • FEN-1 (Flap endonuclease 1), also known as DNase IV, MF1, or RAD2, is a structure-specific nuclease with both 5'-flap endonuclease and 5'-3' exonuclease activities.
  • Plays a critical role in DNA replication by cleaving 5'-overhanging flap structures generated during Okazaki fragment processing, leaving a nick for ligation.
  • Participates in the long patch base excision repair (LP-BER) pathway, cleaving within AP site-terminated flaps.
  • Acts as a genome stabilization factor by preventing flaps from equilibrating into structures that could cause duplications and deletions.
  • Also exhibits 5'-3' exonuclease activity on nicked or gapped double-stranded DNA and possesses RNase H activity.
  • Involved in replication and repair of ribosomal DNA (rDNA) and mitochondrial DNA.
  • Subcellular localization is predominantly nuclear, including the nucleolus and nucleoplasm, but mitochondrial localization has also been reported.
  • Post-translational modifications include acetylation, methylation, and phosphorylation, which may regulate its function and interactions.

  • The immunogen spans amino acids 50-380 of human FEN-1, covering the majority of the protein, which may contribute to recognizing multiple epitopes.
  • For Western blot (WB), expect a band at approximately 43 kDa; consider using whole cell lysates from human or mouse cell lines.
  • For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), nuclear staining is anticipated, but mitochondrial localization may also be observed; optimize fixation and permeabilization conditions.
  • In immunoprecipitation (IP) and ELISA, the polyclonal nature may enhance capture efficiency; validate with appropriate positive and negative controls.
