Biological Background: FDXR Function and Localization
- FDXR encodes NADPH:adrenodoxin oxidoreductase, a mitochondrial flavoprotein also known as ferredoxin reductase. It belongs to the ferredoxin--NADP(+) reductase family and is involved in electron transfer processes.
- The primary function of FDXR is to serve as the first electron transfer protein in all mitochondrial P450 systems, including cholesterol side chain cleavage in steroidogenic tissues, steroid 11-beta hydroxylation in the adrenal cortex, 25-OH-vitamin D3-24 hydroxylation in the kidney, and sterol C-27 hydroxylation in the liver.
- FDXR acts as a ferredoxin--NADP(+) reductase essential for coenzyme Q biosynthesis by transferring electrons to FDX2, which are required for the hydroxylation reaction performed by COQ6. Related references: PMID:38425362
- The protein is localized to the mitochondrion inner membrane, where it mediates electron transport from NADPH to adrenodoxin.
