  • Adrenodoxin (FDX1/ADX), also known as adrenal ferredoxin or hepatoredoxin, is a mitochondrial [2Fe-2S] ferredoxin that serves as an essential electron transfer protein for cytochrome P450 enzymes involved in steroid hormone biosynthesis.
  • Essential for the synthesis of various steroid hormones; transfers electrons from adrenodoxin reductase to CYP11A1, which catalyzes the side-chain cleavage of cholesterol to pregnenolone — the rate-limiting step of steroidogenesis.

  • Does not form a ternary complex with adrenodoxin reductase and CYP11A1 but instead functions as a mobile shuttle, sequentially interacting with each enzyme to transfer electrons (By similarity).
  • Localizes to the mitochondrial matrix, consistent with its role in mitochondrial steroidogenic reactions.
  • Highest expression is detected in the adrenal gland (at protein level); also present in kidney and testis.
