Biological Background: FDX1/ADX Function and Localization
- FDX1, also known as adrenodoxin or adrenal ferredoxin, is a mitochondrial [2Fe-2S] ferredoxin that plays a central role in steroid hormone biosynthesis.
- Functions as an electron transfer shuttle: transfers electrons from adrenodoxin reductase to CYP11A1 (cholesterol side-chain cleavage enzyme), a critical step in the production of all steroid hormones. Related references: PMID:20547883 PMID:21636783
- Localizes to the mitochondrial matrix, consistent with its role in mitochondrial cytochrome P450 reactions.
- Highly expressed in the adrenal gland; also detected at lower levels in kidney and testis.
- Contains essential iron-sulfur and metal-binding domains; post-translational modifications include acetylation and phosphorylation.
- Involved in cholesterol and lipid metabolism through its electron transport function in steroidogenesis.
Experimental Guidance and Technical Tips
- The immunogen is a peptide spanning residues 1–100 of human FDX1/ADX (N-terminal region). This region is exposed in the mature protein and may be conserved across species, consistent with the observed cross-reactivity to mouse and rat.
- For Western blotting, a band near 19 kDa is expected. Optimize blocking conditions and antibody dilution using appropriate positive controls (e.g., adrenal gland or steroidogenic cell lysates).
- For immunofluorescence and IHC, consider mitochondrial co-localization markers. Paraffin-embedded sections may require antigen retrieval; validate staining in tissues with known high FDX1 expression.
- For ELISA applications, pairing with a suitable capture/detection antibody may be required. Optimize coating and detection conditions for your specific assay format.
