Biological Background: FCRL3 Function and Localization
- FCRL3, also known as MAIA, FCRH3, IFGP3, IRTA3, SPAP2, or CD307c, is a single-pass type I membrane protein belonging to the immunoglobulin receptor superfamily. The protein contains multiple immunoglobulin-like domains and a cytoplasmic tail with potential signaling motifs.
- It promotes TLR9-induced B-cell proliferation, activation, and survival but inhibits antibody production and suppresses plasma cell differentiation. FCRL3 enhances activation of NF-κB and MAPK signaling pathways in TLR9-stimulated B-cells. Related references: PMID:23857366
- FCRL3 has inhibitory potential on B-cell receptor (BCR)-mediated signaling, possibly through association with SH2 domain-containing phosphatases. It inhibits cell tyrosine phosphorylation, calcium mobilization, and activation-induced cell death induced through BCR signaling. Related references: PMID:19843936
- Regulatory T-cells expressing FCRL3 exhibit a memory phenotype, are relatively nonresponsive to antigenic stimulation in the presence of IL2, and have a reduced capacity to suppress the proliferation of effector T-cells. Related references: PMID:19494275 PMID:20190142
- FCRL3 acts as a human-specific oocyte surface epitope (MAIA) crucial for sperm-egg adhesion and fusion. It interacts with the IZUMO1-JUNO complex, replacing JUNO as the IZUMO1 receptor during fertilization, thereby permitting species-specific gamete fusion. Related references: PMID:36070373
- Primarily expressed in secondary lymphoid tissues by mature B-cell subsets, with low expression on transitional B-cells and higher on memory B-cells. Also detected in NK cells and a population of thymically derived regulatory T-cells. Expressed in unfertilized oocytes and various tissues including spleen, lymph node, and thymus. - Localized to the cell membrane and enriched on microvillus membrane. Post-translational modifications include glycosylation and phosphorylation, contributing to its molecular weight of approximately 81 kDa.

- For WB, the predicted molecular weight is 81 kDa, but glycosylation may cause a shift to a higher apparent mass. Validate in the relevant sample system with appropriate positive and negative controls.
