Biological Background: FCRL3 Function and Localization

  • FCRL3, also known as MAIA, FCRH3, IFGP3, IRTA3, SPAP2, or CD307c, is a single-pass type I membrane protein belonging to the immunoglobulin receptor superfamily. The protein contains multiple immunoglobulin-like domains and a cytoplasmic tail with potential signaling motifs.
  • It promotes TLR9-induced B-cell proliferation, activation, and survival but inhibits antibody production and suppresses plasma cell differentiation. FCRL3 enhances activation of NF-κB and MAPK signaling pathways in TLR9-stimulated B-cells. Related references: PMID:23857366
  • FCRL3 has inhibitory potential on B-cell receptor (BCR)-mediated signaling, possibly through association with SH2 domain-containing phosphatases. It inhibits cell tyrosine phosphorylation, calcium mobilization, and activation-induced cell death induced through BCR signaling. Related references: PMID:19843936

  • Primarily expressed in secondary lymphoid tissues by mature B-cell subsets, with low expression on transitional B-cells and higher on memory B-cells.
