Biological Background: Ficolin-1 Function and Localization

  • Ficolin-1 (also known as M-ficolin, ficolin-A, ficolin-alpha) is a 35 kDa secreted collagen-like lectin encoded by the FCN1 gene (GeneID: 2219) belonging to the ficolin family of pattern recognition molecules.
  • Acts as a pattern recognition receptor that binds specific carbohydrate structures on pathogen surfaces to initiate the lectin complement pathway via MASP1 serine protease activation, leading to pathogen phagocytosis and adaptive immune signaling.   • For Western blot, reduce samples with DTT/β-mercaptoethanol to detect the 35 kDa monomeric form; note that disulfide-linked oligomers may appear at higher molecular masses under non-reducing conditions.
  • Validate specificity in known FCN1-expressing samples (e.g., human peripheral blood mononuclear cells, U937 or THP-1 monocytic lines) and use a negative control (lymphocytes or knockdown) to confirm the band of interest.
  • In ELISA, conduct checkerboard titrations using recombinant ficolin-1 or serum to optimize coating and detection antibody concentrations; consider testing the antibody as both capture and detection components.
