Biological Background: FCGRT/FCRN Function and Localization
- FCGRT/FCRN, also known as the neonatal Fc receptor (FcRn) or IgG receptor FcRn large subunit p51, mediates the transfer of passive humoral immunity by binding to the Fc region of monomeric IgG and facilitating its selective uptake from maternal milk across the intestinal epithelium (PubMed:10933786, PubMed:7964511).
Related references: PMID:10933786, PMID:7964511 - Throughout life, FcRn contributes to humoral immunity by recycling IgG and extending its half-life; it binds IgG in acidic endosomes and recycles it to the cell surface, releasing it at neutral pH (PubMed:10998088).
Related references: PMID:10998088 - Beyond IgG, FcRn regulates homeostasis of serum albumin (ALB) by a similar recycling mechanism (PubMed:24469444, PubMed:28330995).
Related references: PMID:24469444, PMID:28330995 - FcRn localizes to the cell membrane and endosome membrane, consistent with its role in transcytosis and recycling.
- It is expressed in a wide range of tissues including full-term placenta, heart, lung, liver, muscle, kidney, pancreas, and both fetal and adult small intestine.
- (Microbial infection) FcRn acts as an uncoating receptor for several echoviruses (e.g., Echovirus 5, 6, 7, 9, 11, 13, 25, 29), facilitating infection.
- Key structural features include an immunoglobulin domain, signal peptide, transmembrane helix, disulfide bonds, and N-linked glycosylation; it is also phosphorylated (UniProt keywords: Glycoprotein, Phosphoprotein, Immunoglobulin domain, Disulfide bond).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 27-290), which is expected to produce antibodies that recognize the native receptor conformation; this region encompasses the IgG-binding interface.
- For western blotting, the predicted molecular weight of FcRn is approximately 40 kDa; standard SDS-PAGE and PVDF/nitrocellulose membrane transfer conditions should be optimized empirically.
- IHC-P may benefit from heat-induced epitope retrieval and detection in FcRn-rich tissues such as placenta or small intestine; staining patterns should be confirmed with appropriate controls.
- Given its pH-dependent ligand binding, ELISA assays may require acidic buffers (e.g., pH 6.0) to facilitate FcRn-IgG complex formation; consider assay conditions accordingly.
- The reported cross-reactivity with mouse and rat makes this antibody a candidate for translational studies; however, cross-species performance should be validated in each model system.
