Biological Background: FCGR3B Function and Localization
- Encoded by the FCGR3B gene, also known as CD16b, Fc-gamma RIII-beta, or FcRIIIb, FCGR3B is a low-affinity receptor for the Fc region of immunoglobulin gamma (IgG).
- Unlike the highly related FCGR3A (CD16a), FCGR3B is anchored to the cell membrane via a glycosylphosphatidylinositol (GPI) anchor and lacks a transmembrane domain, consistent with its inability to mediate antibody-dependent cellular cytotoxicity (ADCC) or phagocytosis.
- FCGR3B binds both complexed/aggregated IgG and monomeric IgG, and is thought to function primarily as a trap for immune complexes in the peripheral circulation, preventing aberrant neutrophil activation.
- The protein is specifically expressed on the surface of polymorphonuclear leukocytes (neutrophils) and, to a lesser extent, on stimulated eosinophils, with a secreted form also detected.
- Mature FCGR3B contains two extracellular immunoglobulin-like domains stabilized by disulfide bonds and is heavily glycosylated, contributing to a molecular weight of approximately 26 kDa.

- For Western blotting, anticipate a band near 26 kDa under reducing conditions; however, extensive glycosylation and potential dimerization may lead to higher apparent molecular weights; validate using neutrophil or transfected cell lysates.
- In ELISA applications, consider using the antibody for either capture or detection of recombinant or native FCGR3B; cross-reactivity with rat FCGR3B expands utility in rodent models, but individual experimental titration is recommended.
