Biological Background: FCGR3A Function and Localization
- FCGR3A (CD16a) is a low-affinity transmembrane receptor for the Fc region of immunoglobulin G (IgG), belonging to the immunoglobulin superfamily. It is also known as Fc-gamma RIII-alpha and is a glycoprotein with immunoglobulin-like domains and disulfide bonds.
- Functions as a receptor for clustered antigen–IgG immune complexes, triggering antibody-dependent cellular cytotoxicity (ADCC). It does not bind free monomeric IgG, thereby preventing inappropriate effector cell activation. Related references: PMID:11711607 PMID:21768335 PMID:22023369
- Expressed on natural killer (NK) cells, mediates IgG effector functions including cytokine production and degranulation in response to infection, contributing to the generation of memory-like adaptive NK cells and regulating NK cell survival and proliferation. - For Western blot, a band near 29 kDa is expected. Validate detection using relevant cell lines known to express FCGR3A, such as NK cells, and optimize loading controls.
