Biological Background: FBXW11 Function and Localization

  • FBXW11 (also known as β-TrCP2, Hos, BTRC2, FBW1B) is a member of the F-box/WD repeat protein family, encoded by the FBXW11 gene (GeneID: 23291). It serves as the substrate recognition component of the SCF (SKP1-CUL1-F-box) E3 ubiquitin ligase complex.
  • SCF(FBXW11) mediates the ubiquitination of target proteins by recognizing phosphorylated destruction motifs (D-S-G-X(2,3,4)-S), leading to their proteasomal degradation. Related references: PMID:10437795, PMID:10648623, PMID:11158290, PMID:19966869
  • FBXW11 plays a critical role in Wnt signaling by targeting phosphorylated β-catenin (CTNNB1) for ubiquitin-mediated degradation. Related references: PMID:10321728
  • In the NF-κB pathway, FBXW11 promotes the degradation of phosphorylated IκBα (NFKBIA) and processes the p105 precursor (NFKB1), enabling nuclear translocation and transcriptional activation. Related references: PMID:10321728, PMID:10437795, PMID:10644755, PMID:11158290
  • Additional substrates of SCF(FBXW11) include IFNAR1, CEP68, CDC25A, PER1, PER2, DEPTOR, TFE3, MITF, RCAN1, and CYTH1/2, linking FBXW11 to interferon responses, centriole separation, cell cycle checkpoints, circadian rhythms, mTORC1 signaling, and oxidative stress pathways. Related references: PMID:14532120, PMID:15337770, PMID:25503564, PMID:14603323, PMID:15917222, PMID:22017875, PMID:22017876, PMID:36608670, PMID:18575781, PMID:29420262
  • FBXW11 localizes to both the cytoplasm and nucleus.
  • FBXW11 is implicated in host-virus interactions; for instance, HIV-1 VPU exploits SCF(FBXW11) to ubiquitinate and deplete the antiviral factor BST2. Keywords associated with FBXW11 include alternative splicing, biological rhythms, cell cycle, disease variants, and the Ubl conjugation pathway.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to residues 443-542 of human FBXW11. When using this antibody in mouse or rat samples, confirm epitope conservation and validate cross-reactivity with appropriate controls.
  • For western blotting, the predicted molecular weight of FBXW11 is ~62 kDa; however, observe that post-translational modifications or protein complex formation may affect migration. Include a molecular weight marker and appropriate loading controls.
  • For immunofluorescence/immunocytochemistry (IF/ICC), optimize fixation and permeabilization protocols based on your cell type and target localization (cytoplasmic and nuclear). Use appropriate secondary antibody controls to minimize background.
  • In ELISA applications, determine optimal antibody dilution by titration and consider pairing with a matched detection antibody. Validate assay specificity using antigen competition or knockout controls where possible.
  • As a polyclonal antibody generated in rabbit, lot-to-lot variability may occur; always include positive and negative control samples to ensure consistent performance.
