Biological Background: FBXO7 Function and Localization

  • FBXO7 (F-box only protein 7), also known as FBX, FBX7, PKPS, PARK15, is a substrate recognition component of the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complex, playing roles in cell cycle, proliferation, and chromosome stability. Related references: PMID:15145941 PMID:34791250
  • It mediates ubiquitination and proteasomal degradation of target proteins including BIRC2 and DLGAP5. Related references: PMID:15145941 PMID:16510124 PMID:22212761
  • FBXO7 functions in mitophagy downstream of PINK1, targeting PRKN to damaged mitochondria and promoting MFN1 ubiquitination.
  • It inhibits NF-kappa-B signaling by promoting ubiquitination of UXT isoform 2 and TRAF2. Related references: PMID:33010352 PMID:22212761
  • FBXO7 regulates proteasome assembly and activity by ubiquitinating the PSMA2 subunit with Lys-63-linked polyubiquitin chains.
  • It promotes Lys-48-linked polyubiquitination of SIRT7, leading to hydrogen peroxide-induced cell death. Related references: PMID:36646384
  • FBXO7 localizes to the cytoplasm, nucleus, mitochondrion, and cytosol.
  • Mutations in FBXO7 are associated with Parkinson disease 15 (PARK15), and the protein is linked to neurodegeneration.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (aa 353-522) of human FBXO7. This region may detect full-length protein and some isoforms; confirm specificity using appropriate controls.
  • For Western blotting (WB), a band around 59kDa is expected; however, post-translational modifications may cause apparent molecular weight shifts.
  • Polyclonal antibodies may exhibit lot-to-lot variability; consider titrating the antibody for each new lot and application.
  • The antibody is reported to detect human, mouse, and rat samples; for other species, in-silico sequence alignment can guide cross-reactivity assessment.
  • For immunofluorescence (IF/ICC), optimize fixation and permeabilization conditions.
  • For ELISA, pair with a suitable secondary antibody and validate using recombinant protein or cell lysates.
  • Due to the role of FBXO7 in mitophagy, consider stimulation protocols to induce mitochondrial damage for functional studies.
