Biological Background: FBXO32 Function and Localization

  • Encoded by the FBXO32 gene, F-box only protein 32 (also known as Atrogin-1 and muscle atrophy F-box protein, MAFbx) is a 42 kDa member of the F-box protein family. Multiple isoforms are produced by alternative splicing.
  • FBXO32 functions as the substrate recognition component of an SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complex, mediating the ubiquitination and subsequent proteasomal degradation of target proteins during skeletal muscle atrophy.
  • It specifically recognizes and binds to phosphorylated target proteins, playing a central role in muscle protein degradation pathways associated with atrophy.
  • The protein negatively regulates macrophage efferocytosis by promoting ubiquitination of the transcription factor KLF4, thereby suppressing transcription of the receptor tyrosine kinase MERTK (By similarity).
  • FBXO32 regulates LPS-induced apoptosis and mitochondrial dysfunction through ubiquitination and degradation of ANXA1 (By similarity).
  • It also mediates cyclin D1 protein stabilization via 'Lys-27'-linked ubiquitination. Related references: PMID:40307251
  • Subcellularly, FBXO32 is found in both the cytoplasm and nucleus, consistent with its roles in protein turnover and gene regulation.
  • Expression is highly specific to cardiac and skeletal muscle, underscoring its specialized function in muscle tissue dynamics.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to amino acids 201-300 of human FBXO32; epitope recognition may be influenced by protein conformation or post-translational modifications in different sample preparations.
  • For immunofluorescence (IF/ICC and IF-P) applications, consider appropriate fixation and permeabilization conditions to preserve endogenous FBXO32 localization in muscle cell lines or tissue sections.
  • ELISA assays can be designed with appropriate blocking and detection reagents, and it is advisable to validate the antibody's performance in the relevant sample system using a control antigen.
  • When working with mouse or rat samples, confirm cross-reactivity and signal specificity under your experimental conditions due to possible species differences in the target sequence.
