- FBXO32 (F-box only protein 32), also known as Atrogin-1 or muscle atrophy F-box protein (MAFbx), is the substrate recognition component of the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complex.
- It specifically marks phosphorylated targets, such as TERF1, for ubiquitination and proteasomal degradation, playing a key role in skeletal muscle atrophy.
- FBXO32 negatively regulates macrophage efferocytosis by ubiquitinating the transcription factor KLF4, thereby suppressing MERTK receptor tyrosine kinase transcription (By similarity).
- It regulates lipopolysaccharide (LPS)-induced apoptosis and mitochondrial dysfunction through the ubiquitination and degradation of annexin A1 (ANXA1) (By similarity).
- FBXO32 mediates cyclin D1 protein stabilization via 'Lys-27'-linked ubiquitination. Related references: PMID:40307251
- Primarily localized in the cytoplasm and nucleus, with specific expression in cardiac and skeletal muscle.
