Biological Background: FBXO3 Function and Localization

  • FBXO3 (F-box only protein 3) is the substrate recognition component of the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin ligase complex, targeting proteins such as HIPK2 and EP300 for ubiquitination and proteasomal degradation. Related references: PMID:18809579 PMID:26037928
  • In the presence of PML, FBXO3-mediated ubiquitination of HIPK2 does not lead to immediate degradation, and may synergistically promote p53/TP53-dependent transactivation. Related references: PMID:18809579
  • FBXO3 regulates inflammatory responses by mediating LPS-induced ubiquitination and degradation of FBXL2, specifically recognizing FBXL2 phosphorylated at Thr-404. Related references: PMID:27010866
  • During viral infection, the Rift Valley fever virus NSs protein associates with FBXO3 to form a filamentous E3 ligase that targets the TFIIH complex for degradation, thereby inhibiting antiviral immunity.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1-240) of human FBXO3. This region may be conserved across isoforms; therefore, the antibody could detect multiple splice variants. Validate in relevant human cell lysates.
  • For Western blot, the predicted molecular weight is approximately 55 kDa. Include appropriate positive controls, such as HeLa or HEK293T cell lysates. Due to reported cross-reactivity with mouse, mouse tissue or cell lysates may also be tested.
  • In ELISA applications, use recombinant FBXO3 protein as a standard and optimize coating and detection conditions empirically.
  • The antibody may be suitable for studies on ubiquitin-mediated proteolysis, inflammation, or viral pathogenesis; however, specific performance should be confirmed in the intended experimental system.
