Biological Background: FBXO22 Function and Localization
- FBXO22 (F-box only protein 22, also known as FBX22 or FISTC1) is a substrate-recognition component of the SCF E3 ubiquitin ligase complex involved in ubiquitin-mediated proteolysis.
- It promotes degradation of sarcomeric proteins alpha-actinin (ACTN2) and filamin-C (FLNC), which is critical for cardiac and skeletal muscle contractile function.
- FBXO22 controls histone H3K9 and H3K36 methylation by regulating the stability of the histone demethylase KDM4A. Related references: PMID:21768309
- Through KDM4A, it targets methylated TP53 for degradation, influencing cellular senescence. Related references: PMID:26868148
- Under oxidative stress, FBXO22 ubiquitinates and promotes degradation of BACH1, relieving transcriptional repression. Related references: PMID:39504958
- It mediates 'Lys-27'-linked ubiquitination of MTOR upon amino acid starvation, inhibiting mTORC1 signaling. Related references: PMID:37979583
- FBXO22 inhibits SARS-CoV-2 replication by targeting viral NSP5 for degradation. Related references: PMID:39223933
- Subcellularly, it localizes to the cytoplasm, nucleus, and sarcomere Z-line; predominantly expressed in liver and cardiac muscle.
Experimental Guidance and Technical Tips
- For Western blot, expect a band at approximately 45 kDa; validate using lysates from human cell lines or tissues known to express FBXO22 (e.g., liver, cardiac muscle).
- The immunogen spans amino acids 104–403, covering a region that may detect multiple isoforms or splice variants; consider this when interpreting band patterns.
- In ELISA, optimize antigen coating (recombinant protein or peptide) and antibody dilution using a standard curve for quantitation.
- Due to the polyclonal nature, lot-to-lot variability can occur; always validate in your specific assay system.
