Biological Background: FBXO2 Function and Localization

  • FBXO2 (F-box only protein 2), also known as FBG1, FBX2, Fbs1, OCP1, or NFB42, is a member of the F-box protein family and functions as the substrate recognition subunit of the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin ligase complex.
  • It specifically recognizes and binds high-mannose-type sugar chains on misfolded glycoproteins that have been retrotranslocated from the endoplasmic reticulum (ER) to the cytosol, targeting them for ubiquitination and proteasomal degradation as part of the ER-associated degradation (ERAD) pathway.
  • By promoting the clearance of unfolded glycoproteins, FBXO2 helps prevent the formation of potentially toxic cytosolic protein aggregates, maintaining cellular proteostasis.
  • FBXO2 localizes predominantly to the cytoplasm and is associated with microsomal membranes, consistent with its role in ERAD.
  • As a lectin-type F-box protein, it is implicated in the ubiquitin-proteasome system and has been identified through proteomics studies. No tissue-specific or antimicrobial functions are reported.

Experimental Guidance and Technical Tips

  • The immunogen is a recombinant protein covering the full-length human FBXO2 (amino acids 1–296), which is expected to generate polyclonal antibodies recognizing multiple epitopes across the protein.
  • Given the high sequence homology among mammalian FBXO2 orthologs, the antibody has demonstrated cross-reactivity with mouse and rat samples; consider validating in your specific experimental system.
  • The antibody is suitable for western blot (WB) and ELISA; for optimal results, titrate the antibody concentration and use appropriate positive controls expressing FBXO2.
  • For WB, note that the calculated molecular weight of FBXO2 is approximately 33 kDa; confirm band specificity using knockout or knockdown controls when possible.
