Biological Background: FBXL5 Function and Localization

  • FBXL5 (F-box/LRR-repeat protein 5) is a component of the SCF (SKP1-cullin-F-box) E3 ubiquitin-protein ligase complex. It regulates iron homeostasis by targeting IREB2/IRP2 for ubiquitination and degradation. Related references: PMID:19762596, PMID:19762597
  • The N-terminal hemerythrin-like (Hr) domain contains a diiron metal center that acts as an iron sensor. Under low iron, FBXL5 is unstable; iron binding stabilizes the protein. Related references: PMID:19762596, PMID:19762597
  • The C-terminal domain contains a redox-sensitive [2Fe-2S] cluster, which upon oxidation enables FBXL5 to recruit IRP2 for degradation in an oxygen-dependent manner. Related references: PMID:32126207

  • The immunogen covers amino acids 1–310, corresponding to the N-terminal hemerythrin-like and F-box domains, which are evolutionarily conserved across human, mouse, and rat. This region likely accounts for the observed cross-reactivity.
  • For western blotting (WB), consider the theoretical molecular weight of approximately 79 kDa (UniProt) or the supplier-reported 79 kDa. Confirm band size in your specific sample matrix and validate signal specificity using appropriate controls (e.g., knockdown or knockout lysates).
  • In ELISA applications, the polyclonal nature may allow detection of both native and denatured target protein. Optimize coating and detection conditions empirically.
  • Note that FBXL5 undergoes alternative splicing; therefore, multiple bands may be detected in WB. Validate the antibody’s reactivity profile in the relevant tissue or cell line.
