  • F-box only protein 32 (FBXO32), also known as Atrogin-1 or muscle atrophy F-box protein (MAFbx), is encoded by the FBXO32 gene and plays a pivotal role in ubiquitin-mediated proteolysis.
  • As a substrate recognition component of the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complex, FBXO32 mediates the ubiquitination and subsequent proteasomal degradation of target proteins, particularly during skeletal muscle atrophy.
  • FBXO32 specifically recognizes and binds to phosphorylated target proteins, facilitating their degradation and thereby regulating muscle mass.
  • Subcellularly, FBXO32 localizes to both the cytoplasm and nucleus, consistent with its role in protein turnover across cellular compartments.
  • FBXO32 is specifically expressed in cardiac and skeletal muscle, underscoring its tissue-specific function in muscle biology.
  • Beyond muscle atrophy, FBXO32 negatively regulates macrophage efferocytosis by promoting ubiquitination of KLF4, and regulates LPS-induced apoptosis and mitochondrial dysfunction through ANXA1 ubiquitination (By similarity).
