- Fatty acid synthase is a multi-enzyme protein that catalyzes the de novo biosynthesis of palmitate, a long-chain saturated fatty acid, from acetyl-CoA and malonyl-CoA in the presence of NADPH. It harbors seven distinct catalytic activities and a 4'-phosphopantetheine prosthetic group binding site (acyl carrier protein domain).
- The enzyme plays a central role in lipid biosynthesis and is essential for membrane synthesis and energy storage. Its activity is tightly linked to cellular metabolic states, and it is frequently upregulated in cancer and metabolic disorders.
- FASN localizes predominantly to the cytoplasm and has also been detected in melanosomes, where its activity contributes to melanin synthesis.
- It is ubiquitously expressed, with particularly high levels in brain, lung, liver, and mammary gland, reflecting its importance in lipogenic tissues.

Experimental Guidance and Technical Tips
- Due to the large molecular weight of FASN (~273 kDa), consider using low-percentage SDS-PAGE gels (e.g., 6% or gradient gels) and extended wet transfer times for Western blotting. A prestained high-molecular-weight marker is recommended.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval with citrate buffer (pH 6.0) or EDTA buffer (pH 9.0) may be required. Verify optimal conditions in your tissue and cell line models.
- In ELISA applications, use the recombinant immunogen as a positive control. Titrate both the capture and detection antibodies to establish working concentrations.
