Biological Background: FAT10/UBD Function and Localization

  • FAT10 (Ubiquitin D) is a ubiquitin-like protein modifier that can be covalently conjugated to target proteins, targeting them for degradation by the 26S proteasome in a NUB1-dependent manner. Related references: PMID:15831455, PMID:16707496, PMID:19166848
  • Conjugation is activated by the E1-like enzyme UBA6 through adenylation of the C-terminal glycine. Related references: PMID:17889673, PMID:35970836
  • FAT10 promotes the expression of the immunoproteasome subunit PSMB9/LMP2 and regulates NF-κB activation by mediating TNF-induced proteasomal degradation of ubiquitinated IκBα. Related references: PMID:19959714
  • Involved in dendritic cell maturation and constitutively expressed in mature dendritic cells and B cells, as well as in tissues of the reticuloendothelial system, gastrointestinal system, kidney, lung, and prostate. Related references: PMID:19028597
  • Mediates mitotic non-disjunction and chromosome instability by impairing the kinetochore localization of MAD2L1 during prometaphase. Related references: PMID:16495226
  • Under conditions of proteasome saturation or impairment, FAT10 is involved in the formation of aggresomes. Related references: PMID:19033385
  • FAT10 can induce caspase-dependent apoptosis, particularly in renal epithelium and tubular cells, contributing to kidney diseases like polycystic kidney disease and HIV-associated nephropathy. Related references: PMID:16495380
  • Subcellularly localized to both the nucleus and cytoplasm, FAT10 undergoes acetylation and participates in the ubiquitin-like conjugation pathway.

Experimental Guidance and Technical Tips

  • The immunogen used is a recombinant fusion protein corresponding to amino acids 1-165 of human FAT10/UBD. Consider this when designing epitope mapping or antibody characterization experiments.
  • For Western blotting, the predicted protein weight is 18 kDa; use appropriate molecular weight markers and loading controls in this range.
  • In immunofluorescence/immunocytochemistry (IF/ICC), optimize fixation/permeabilization conditions to preserve the nuclear and cytoplasmic localization of FAT10/UBD.
  • This antibody shows cross-reactivity with mouse FAT10/UBD, making it suitable for studies in mouse models, but in-species validation is recommended.
  • For ELISA applications, perform standard checkerboard titrations to determine optimal coating antibody and detection conditions.
