Biological Background: FAT10/UBD Function and Localization

  • FAT10/UBD (ubiquitin D) is a ubiquitin-like protein modifier consisting of two tandem ubiquitin-like domains, leading to the alternative name “diubiquitin.”
  • Conjugated to target proteins via UBA6-mediated adenylation of its C-terminal glycine, it tags them for 26S proteasomal degradation in a NUB1-dependent manner.

  • Positively regulates NF-kappa-B signaling by promoting TNF-mediated proteasomal degradation of ubiquitinated I-kappa-B-alpha, and is required for TNF-induced p65 nuclear translocation in renal tubular epithelial cells.
