Biological Background: FAT10/UBD Function and Localization

  • FAT10/UBD (Ubiquitin D), also known as diubiquitin, is a ubiquitin-like protein that functions as a covalent modifier targeting substrates for 26S proteasomal degradation in an NUB1-dependent manner.   • It positively regulates NF-κB signaling by promoting the TNF-induced proteasomal degradation of ubiquitinated IκBα. Related references: PMID:19959714

Experimental Guidance and Technical Tips

  • The antibody was raised against a synthetic peptide covering amino acids 70–150 of human FAT10/UBD. Consider using this region-specific design to minimize cross-reactivity with other ubiquitin-like proteins.
  • For Western blot, test on whole-cell lysates from cell lines known to express FAT10, such as human B-cell or dendritic cell lines, or upon TNF/LPS stimulation to induce endogenous expression. Mouse cell lines may also be used given confirmed cross-reactivity.
  • In ELISA, the antibody may serve as a capture or detection reagent for quantifying FAT10 levels. Validate assay conditions with recombinant FAT10 protein or cell extracts.
