Biological Background: FASTKD2 Function and Localization

  • FASTKD2 (FAST kinase domain-containing protein 2) is a mitochondrial protein encoded by the FASTKD2 gene, also known as COXPD44 or KIAA0971.
  • It plays an important role in the assembly of the mitochondrial large ribosomal subunit by functioning as part of a protein-RNA module that controls 16S mitochondrial ribosomal RNA (mt-rRNA) abundance, which is required for intra-mitochondrial translation.   • May also play a role in mitochondrial apoptosis.
  • Subcellular localization is primarily in the mitochondrion matrix and nucleoid.
  • Expression is highest in heart, smooth muscle, and thyroid, with detection in a wide range of tissues including spleen, thymus, testis, ovary, colon, kidney, brain, lung, and liver.
  • Post-translational modifications include phosphorylation (phosphoprotein), and the protein undergoes alternative splicing.
  • Variants in FASTKD2 are associated with primary mitochondrial disease, specifically combined oxidative phosphorylation deficiency 44 (COXPD44).
