Biological Background: FAP Function and Localization

  • Prolyl endopeptidase FAP (Fibroblast activation protein alpha, also known as seprase) is a cell surface glycoprotein serine protease involved in extracellular matrix (ECM) degradation, contributing to tissue remodeling, fibrosis, wound healing, inflammation, and tumor growth. It exhibits post-proline cleaving endopeptidase activity with a preference for Ala/Ser-Gly-Pro-Ser/Asn/Ala consensus sequences on substrates like SERPINF2 and SPRY2. Related references: PMID:14751930 PMID:16223769 PMID:16410248

  • FAP degrades gelatin and heat-denatured type I collagen, but not native collagens type I and IV, vitronectin, tenascin, laminin, fibronectin, fibrin or casein. Related references: PMID:10347120 PMID:10455171 PMID:12376466

  • The enzyme also possesses dipeptidyl peptidase activity, cleaving N-terminal dipeptides where the penultimate residue is proline, with a preference for Ala-Pro, Ile-Pro, Gly-Pro, Arg-Pro, and Pro-Pro. Related references: PMID:10347120 PMID:10593948 PMID:16175601

  • Natural substrates for the dipeptidyl peptidase activity include neuropeptide Y, peptide YY, substance P, and brain natriuretic peptide 32. Related references: PMID:21314817

  • The plasma membrane form of FAP, in association with DPP4, PLAUR or integrins, promotes pericellular proteolysis, cell adhesion, migration, and invasion, thereby enhancing tumor progression, angiogenesis, and immune evasion.

  • FAP localizes to the cell surface, membrane, lamellipodia, invadopodia, and ruffles, and is also secreted and found in the cytoplasm. It is highly expressed in activated fibroblasts of healing wounds, tumor stroma, cirrhotic liver, and adipose tissue, with isoform-specific expression in melanoma, carcinoma, and fibroblast cell lines.

Experimental Guidance and Technical Tips

  • For flow cytometry, titrate the antibody to determine the optimal staining concentration for your specific cell type and experimental conditions.
  • The immunogen spans the extracellular domain (aa 26-760), so the antibody may detect both membrane-bound and soluble FAP; validate detection specificity in your assay system.
  • Use appropriate isotype controls and fix/permeabilize cells if intracellular staining is required.
  • Validate antibody performance in relevant human samples (e.g., activated fibroblasts, tumor cell lines) as cross-reactivity is limited to human.
