Biological Background: FANCM Function and Localization
- FANCM is a DNA-dependent ATPase and integral component of the Fanconi anemia core complex, essential for activating the FA pathway and promoting FANCI-FANCD2 monoubiquitination upon DNA damage. Related references: PMID:16116422 PMID:19423727 PMID:20347428
- In complex with CENPS/CENPX, FANCM binds dsDNA, fork-structured DNA, and Holliday junctions, and its ATP-dependent branch migration activity remodels replication forks; with FAAP24, it preferentially binds ssDNA and splayed-arm substrates. Related references: PMID:20347428 PMID:20347429 PMID:17289582 PMID:16116434
- Localized in the nucleus, consistent with its role in DNA repair and genome maintenance.
- Expressed in germ cells of fetal and adult ovaries, including oogonia and pachytene oocytes, and in the testis. Related references: PMID:29231814 PMID:30075111
- Belongs to the helicase superfamily, with conserved ATP-binding and DNA-binding domains; classified as a phosphoprotein involved in DNA damage response and disease variants linked to Fanconi anemia and premature ovarian failure.
- Multiple isoforms exist due to alternative splicing.
Experimental Guidance and Technical Tips
- For Western blot, use whole cell lysates from human cells and consider extended transfer times due to the large 232 kDa protein size.
- In IF/ICC, verify nuclear staining pattern; fixation and permeabilization conditions should be optimized for nuclear antigens.
- ELISA applications may utilize the recombinant immunogen as a positive control; antibody titer and specificity should be validated in the relevant sample matrix.
