Biological Background: TASOR Function and Localization
- Core component of the HUSH complex, a multiprotein complex that specifically mediates epigenetic repression of mobile genetic elements, such as retroviruses and transposable elements. Related references: PMID:26022416, PMID:28581500, PMID:29211708
- Represses LINE-1 (L1) retrotransposons that are still capable of transposition; silencing events often occur within introns of transcriptionally active genes and lead to down-regulation of host gene expression. Related references: PMID:29211708, PMID:32976585
- Acts as the central scaffold within the HUSH complex, recruiting MPHOSPH8, PPHLN1, SETDB1 (a histone methyltransferase depositing H3K9me3), and MORC2 (a chromatin remodeler that compacts chromatin) to mediate gene silencing. Related references: PMID:32976585, PMID:33009411, PMID:39589886
- Mediates silencing of unintegrated retroviral DNA following recruitment by ZNF638; this transcriptional repression occurs immediately after infection before integration into the host genome. Related references: PMID:30487602
- Subcellular localization: predominantly found in the nucleus and associated with chromosomes, consistent with its role in chromatin-based transcriptional repression.
- Post-translational modifications include acetylation, phosphorylation, and isopeptide bond formation; the protein also undergoes alternative splicing, producing multiple isoforms.
- Plays a crucial role in early embryonic development by maintaining epiblast cell fitness and normal cell division (By similarity).
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 700–1000 of human FAM208A (NP_001106207.1). Consider verifying reactivity with known isoforms that may lack this region.
- For Western blot (WB), validate the antibody in human cell lines expressing TASOR, using appropriate positive and negative controls. The predicted molecular weight is ~189 kDa; use an appropriate gel percentage and extended transfer conditions for high-molecular-weight proteins.
- For ELISA, optimize coating conditions and antibody dilution; cross-reactivity with other species has not been tested, so validate in human-derived samples.
- Perform initial titration experiments to determine the optimal working concentration for your specific assay system and sample type.
