Biological Background: FAM107A Function and Localization
- Actin-associated protein FAM107A (also known as DRR1, TU3A) is encoded by the FAM107A gene and undergoes alternative splicing.
- Core function as a stress-inducible actin-binding protein: mediates polymerization of globular actin to F-actin, stabilizes and bundles F-actin, and contributes to synaptic functions such as neurite outgrowth and hippocampus-dependent cognitive processes (By similarity).
- Involved in actin and microtubule cytoskeleton organization; negatively regulates focal adhesion assembly, promoting malignant glial cell migration in an actin-, microtubule- and MAP1A-dependent manner. Related references: PMID:20543869
- Inhibits neuroblastoma G1/S phase cell cycle progression by stimulating ubiquitination and degradation of NF-kappa-B subunit RELA in a COMMD1- and actin-dependent manner. Related references: PMID:10564580, PMID:28604741
- Subcellular localization includes nucleus, cytoplasm, stress fibers, focal adhesions, ruffle membranes, and synapses.
- Widely expressed, notably in neurons and malignant glial tumors; expression is reduced or absent in many cancer cell lines. Related references: PMID:10564580
- May play a role in tumor development. Related references: PMID:10564580
Experimental Guidance and Technical Tips
- For Western blot (WB), the predicted molecular weight is approximately 17 kDa; validate detection in appropriate human or mouse cell/tissue lysates.
- ELISA applications may require optimization of coating antigen concentration and antibody dilution; consider using the recombinant immunogen as a positive control.
- Because the antibody cross-reacts with mouse, it can be utilized for studies involving mouse models, though validation in the specific sample type is recommended.
