Biological Background: FAM107A Function and Localization
- FAM107A (also designated DRR1 or TU3A) is a stress-inducible actin-binding protein that promotes actin polymerization and stabilizes/bundles filamentous actin (F-actin), essential for synaptic functions such as neurite outgrowth and hippocampus-dependent cognitive processes.
- Negatively regulates focal adhesion (FA) assembly to facilitate malignant glial cell migration in an actin-, microtubule-, and MAP1A-dependent manner.
Related references: PMID:20543869 - Inhibits G1/S phase cell cycle progression and cell proliferation by stimulating ubiquitination and degradation of the NF-kappa-B subunit RELA through a COMMD1- and actin-dependent pathway.
Related references: PMID:10564580 PMID:28604741 - May contribute to tumor development, as its expression is frequently downregulated in cancer cell lines.
Related references: PMID:10564580 - Localizes to multiple subcellular compartments, including the nucleus, cytoplasm, stress fibers, focal adhesions, ruffle membranes, and synapses, reflecting its roles in cytoskeletal organization and cell adhesion.
- Widely expressed in normal tissues, with prominent presence in neurons and malignant glial tumors, but reduced or absent in many cancer types.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal 1–144 amino acids of human FAM107A; a band at approximately 17 kDa is expected in Western blot assays on appropriate lysates.
- For ELISA, pairing with a matched detection antibody may improve sensitivity; validate cross-reactivity with mouse and rat samples, as indicated.
- Optimization of antibody dilution in the specific sample system is recommended, as expression of FAM107A can vary among cell lines and tissues.
- Given the multi-compartment localization of FAM107A, consider subcellular fractionation or co-immunostaining to confirm target-specific interactions.
