  • Protein identity: FAK (Focal adhesion kinase 1), also known as PTK2, is a non-receptor protein-tyrosine kinase that serves as a critical regulator of cell adhesion, migration, and cytoskeletal reorganization. It exists in multiple isoforms, including full-length FAK and an inhibitory isoform FRNK (FAK-related non-kinase) that lacks the kinase domain.

  • Gene family: Encoded by the PTK2 gene, FAK belongs to the FAK family of tyrosine kinases. It is ubiquitously expressed, with prominent roles in embryonic development, angiogenesis, and nervous system formation.
  • Subcellular localization: FAK localizes primarily to focal adhesions at the cell membrane and is also found in the cytoplasm, nucleus, centrosome, and cilium basal body. Its shuttling between adhesion sites and the nucleus allows integration of mechanical and growth signals.

Experimental Guidance and Technical Tips

  • The immunogen (amino acids 734–834) corresponds to the C-terminal focal adhesion targeting (FAT) domain, which is present in both full-length FAK and the FRNK isoform. Therefore, the antibody may detect both forms—consider validating in your cell/tissue model with appropriate molecular weight markers (full-length ~119 kDa, FRNK ~41 kDa).
  • For western blotting, ensure sufficient protein loading and optimize primary antibody dilution; a band at 119 kDa is expected for full-length FAK in most human, mouse, and rat samples.
  • In immunohistochemistry and immunofluorescence, FAK typically concentrates at focal adhesions near the cell periphery, but may also show cytoplasmic or perinuclear staining depending on cell type and activation state. Use appropriate fixation and permeabilization methods.
  • For immunoprecipitation, the polyclonal nature may provide robust capture; confirm with positive control lysates.
