Biological Background: FAF2 Function and Localization
- FAF2 (FAS-associated factor 2), also known as UBXD8, is a UBX domain-containing protein encoded by the FAF2 gene in humans.
- It functions in endoplasmic reticulum-associated degradation (ERAD), facilitating ubiquitin-dependent degradation of misfolded ER proteins. Related references: PMID:18711132, PMID:24215460
- FAF2 regulates insulin-like growth factor receptor signaling by controlling IGF1R steady-state expression. Related references: PMID:26692333
- At lipid droplets, FAF2 recruits the ATPase VCP and binds to PNPLA2/ATGL, inhibiting triacylglycerol hydrolysis and reducing lipid droplet degradation. Related references: PMID:23297223
- It participates in stress granule disassembly by associating with ubiquitinated G3BP1, promoting VCP-mediated extraction from stress granules. Related references: PMID:34739333
- FAF2 localizes to the cytoplasm, lipid droplets, and endoplasmic reticulum.
- Broadly expressed with highest levels in brain.
- Post-translational modifications include acetylation; the protein contains a coiled coil domain and is involved in the unfolded protein response.
Experimental Guidance and Technical Tips
- The immunogen covers the middle portion of FAF2 (aa 176-393); consider using this sequence for peptide blocking or epitope mapping studies.
- For Western blot, a band around 53 kDa (endogenous FAF2) may be detected; confirm specificity using positive controls (e.g., HeLa, HepG2) and negative controls (knockdown/knockout).
- In immunofluorescence/ICC, fix and permeabilize cells appropriately; co-staining with organelle markers can verify localization to lipid droplets or ER.
- For ELISA, the antibody can be used as capture or detection reagent; optimize coating and detection conditions.
- Cross-reactivity with mouse and rat suggests conserved epitopes; verify in relevant models.
