Biological Background: FADS3 Function and Localization
- FADS3 (fatty acid desaturase 3), also known as delta(13) desaturase, is a member of the fatty acid desaturase family.
- FADS3 catalyzes the introduction of a cis (Z) double bond between carbon 14 and carbon 15 of the sphingoid base, converting sphingosine (SPH) to sphinga-4,14-dienine (SPD). It exhibits a preference for SPH-containing ceramides as substrates. Related references: PMID:31862735, PMID:31916624, PMID:37209771
- SPD ceramides, generated by FADS3, are widely distributed in mammalian tissues and may function oppositely to conventional ceramides in lipid microdomain formation due to their cis double bond configuration. Related references: PMID:31916624
- The desaturase mechanism of FADS3 relies on electron transfer facilitated by cytochrome b5. Related references: PMID:37209771
- FADS3 also acts as a methyl-end fatty acyl-CoA desaturase, capable of converting trans-vaccenoate to (11E,13Z)-octadecadienoate (a conjugated linoleic acid isomer). (By similarity)
- It may play a role in detoxifying 1-deoxysphingolipids by desaturating cytotoxic 1-deoxysphinganine to 1-deoxysphingenine. (Probable)
- Subcellularly, FADS3 is an endoplasmic reticulum membrane protein with multiple transmembrane domains.
- FADS3 is highly expressed at the protein level in liver, kidney, brain, lung, pancreas, testis, ovary, and skeletal muscle.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-130 of human FADS3, suggesting the antibody likely recognizes the N-terminal region. Consider this when designing epitope-tagging or truncation experiments.
- For Western blot (WB), the predicted molecular weight is 51 kDa; run appropriate molecular weight markers and validate band specificity in the relevant sample system.
- For ELISA, perform titration experiments to determine the optimal coating concentration and detection conditions for your assay format.
- Since the antibody shows cross-reactivity with human, mouse, and rat, it may be suitable for studies across these species; however, validate in the target tissue or cell line of interest.
- As a polyclonal antibody, expect batch-to-batch variability; consider including appropriate positive and negative controls in each experiment.
