Biological Background: FADS1 Function and Localization
- FADS1 (Fatty Acid Desaturase 1) is an acyl-CoA (8-3)-desaturase that introduces a cis double bond at carbon 5 of fatty acyl chains. It is also known as Delta(5) fatty acid desaturase (D5D).
- It plays a key role in the biosynthesis of highly unsaturated fatty acids (HUFAs) from essential polyunsaturated fatty acids (PUFAs) like linoleic acid and alpha-linolenic acid.
- Specifically, it desaturates dihomo-gamma-linoleate (DGLA, 20:3n-6) to arachidonate (AA, 20:4n-6) and eicosatetraenoate (ETA, 20:4n-3) to eicosapentaenoate (EPA, 20:5n-3), a rate-limiting step in eicosanoid production. Related references: PMID:10601301, PMID:10769175
- FADS1 is a membrane-bound enzyme localized primarily to the endoplasmic reticulum and also detected in mitochondria.
- It is widely expressed, with highest levels in liver, brain, adrenal gland, and heart, and is abundant in fetal liver and brain.
- The protein contains several transmembrane helices and is subject to post-translational modifications including acetylation and alternative splicing.
- FADS1 controls the metabolism of inflammatory lipids, such as prostaglandin E2, contributing to acute inflammatory response and epithelial homeostasis.
- It does not exhibit catalytic activity toward 20:3n-6 but may enhance FADS2 activity.
