Biological Background: FADD Function and Localization
- FAS-associated death domain protein (FADD), also known as MORT1 and GIG3, is a critical adapter molecule in death receptor-mediated apoptosis and innate immune signaling.
- Acts as an apoptotic adaptor that recruits caspases CASP8 or CASP10 to activated FAS/CD95 or TNFRSF1A/TNFR-1 receptors, forming the death-inducing signaling complex (DISC) to trigger caspase activation and apoptosis. Related references: PMID:16762833, PMID:19118384, PMID:20935634
- Participates in interferon-mediated antiviral innate immune responses by positively regulating interferon signaling pathways. Related references: PMID:21109225, PMID:24204270
- Localizes predominantly to the cytoplasm, consistent with its role as a signal transduction mediator.
- Widely expressed across human tissues, with the notable exception of peripheral blood mononuclear leukocytes.
- Post-translational modifications include phosphorylation and glycosylation, which may regulate its activity and interactions.
Experimental Guidance and Technical Tips
- For Western blotting, use whole cell lysates from human cell lines (e.g., HeLa or Jurkat) under reducing conditions; a specific band near 23 kDa is expected. Validate with a FADD-positive control.
- In ELISA, the recombinant immunogen (aa 20-150) can serve as a positive coating antigen. Perform checkerboard titrations to optimize antibody and antigen concentrations.
- As FADD is primarily cytoplasmic, subcellular fractionation or immunofluorescence co-staining may improve detection specificity. Include appropriate isotype controls for comparison.
