Biological Background: Fatty acid-binding protein, intestinal Function and Localization
- Encoded by the FABP2 gene, fatty acid-binding protein, intestinal (also known as I-FABP or FABPI) is a ~15 kDa cytosolic lipid-binding protein belonging to the intracellular fatty acid-binding protein family.
- Plays a key role in the intracellular transport of long-chain fatty acids and their acyl-CoA esters, facilitating their movement within the cytoplasm.
- Involved in triglyceride-rich lipoprotein synthesis, contributing to dietary fat absorption and chylomicron assembly in enterocytes.
- Exhibits high binding affinity for saturated long-chain fatty acids and relatively lower affinity for unsaturated long-chain fatty acids.
- Functions as a lipid sensor, potentially helping to maintain energy homeostasis by modulating lipid-mediated signaling.
- Primarily expressed in the small intestine, with highest levels in the jejunum, and much lower expression in the large intestine; restricted to absorptive epithelial cells.
- Post-translationally modified by acetylation; also identified in proteomics studies.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant protein comprising amino acids 1-132 of human FABPI, ensuring recognition of full-length endogenous protein. For immunoblotting, the predicted molecular weight is ~15 kDa; validate band size in relevant tissue lysates (e.g., jejunum or small intestine).
- This polyclonal antibody is suitable for WB and ELISA applications. Consider optimizing dilutions for your specific sample type and detection system. Endogenous expression in intestinal tissues and cross-reactivity with mouse and rat may facilitate translational studies.
- Since FABPI is a cytoplasmic protein, standard whole-cell or tissue lysates can be used. For localization studies, additional fixation or permeabilization steps may be required if extended to ICC/IHC; however, these applications are not validated, so validate accordingly.
