Biological Background: FABPI Function and Localization

  • FABPI, also known as fatty acid-binding protein 2 (FABP2) or intestinal-type fatty acid-binding protein (I-FABP), is a lipid-binding protein primarily expressed in the intestine.
  • It binds saturated long-chain fatty acids with high affinity and unsaturated long-chain fatty acids with lower affinity, playing a role in intracellular fatty acid transport and acyl-CoA ester trafficking.
  • FABP2 is involved in triglyceride-rich lipoprotein synthesis and may function as a lipid sensor to help maintain energy homeostasis.
  • The protein localizes to the cytoplasm and is highly expressed in the small intestine, particularly the jejunum, with lower levels in the large intestine.
  • Post-translational modifications include acetylation, and its 3D structure has been resolved (reference proteome).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1–100) of human FABPI, ensuring target specificity.
  • For Western blot, expect a band at approximately 15 kDa; validate using intestinal tissue lysates, preferably from jejunum where expression is highest.
  • Due to cross-reactivity with mouse and rat, this antibody may be suitable for comparative studies across species.
  • For immunohistochemistry and immunofluorescence, optimize antigen retrieval and detection conditions appropriately.
