Biological Background: FABP7 Function and Localization
- FABP7, also known as brain lipid-binding protein (BLBP), brain-type fatty acid-binding protein (B-FABP), or mammary-derived growth inhibitor related (MRG), is a member of the fatty acid-binding protein family.
- It is thought to be involved in the transport of an unidentified hydrophobic ligand with potential morphogenic activity during central nervous system (CNS) development.
- FABP7 is required for the establishment of the radial glial fiber system in the developing brain, which is essential for the migration of immature neurons and the formation of cortical layers (By similarity).
- The protein is predominantly localized in the cytoplasm.
- It is specifically expressed in brain and other neural tissues.
- Post-translational modifications include acetylation, and alternative splicing yields multiple isoforms.
- The protein has a molecular weight of approximately 15 kDa and is classified under keywords such as lipid-binding, transport, and direct protein sequencing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length sequence (amino acids 1-132) of human FABP7, which is highly conserved in rat. Therefore, cross-reactivity with rat is observed, but reactivity with other species should be validated experimentally.
- For Western blot (WB), a band of approximately 15 kDa is expected; consider optimizing blocking and antibody dilution based on the sample type and endogenous expression levels.
- In immunofluorescence/immunocytochemistry (IF/ICC), localization should appear cytoplasmic, consistent with the subcellular distribution of FABP7. Include appropriate controls to confirm specificity.
- For ELISA, the antibody can be used as a capture or detection reagent; titration and pairing with a suitable secondary antibody are recommended for optimal signal.
