Biological Background: FABP5 Function and Localization
- FABP5 is an intracellular carrier for long-chain fatty acids and related active lipids, such as endocannabinoids, regulating their metabolism and signaling.
- Selectively delivers specific fatty acids from the cytosol to the nucleus, activating nuclear receptors including PPARδ, thereby promoting proliferation and survival. Related references: PMID:21395585 PMID:22170058
- Functions as a synaptic carrier of endocannabinoids at central synapses, controlling retrograde signaling.
- Modulates inflammation by regulating PTGES induction via NF-κB activation and PGE2 biosynthesis.
- Highly expressed in keratinocytes, especially in psoriatic skin, suggesting a role in keratinocyte differentiation. Related references: PMID:8092987
- Localizes to the cytoplasm, nucleus, synapse, postsynaptic density, and is also secreted.
- Undergoes post-translational modifications including acetylation, phosphorylation, and contains disulfide bonds.
- Belongs to the fatty acid-binding protein family and is also known as epidermal-type fatty acid-binding protein (E-FABP).
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering amino acids 1-135 of human FABP5 (full-length), which may contribute to detecting the endogenous protein.
- Based on the immunogen sequence and cross-reactivity data (Human, Mouse, Rat), the antibody likely recognizes FABP5 across these species; validation in each species is advised.
- For WB, consider using a blocking agent compatible with rabbit polyclonal antibodies and validate the expected band at ~15 kDa.
- For IHC-P, tissue fixation and antigen retrieval conditions should be optimized for each sample type.
- For IF/ICC, appropriate fixation and permeabilization methods should be tested.
- ELISA performance can be evaluated with recombinant FABP5 protein or cell lysates.
