Biological Background: FABP5 Function and Localization

  • FABP5, also known as epidermal-type fatty acid-binding protein (E-FABP) or psoriasis-associated fatty acid-binding protein homolog (PA-FABP), is a member of the fatty acid-binding protein family. It functions as an intracellular carrier for long-chain fatty acids and related active lipids, such as endocannabinoids, regulating the metabolism and actions of bound ligands.
  • In addition to cytosolic transport, FABP5 selectively delivers specific fatty acids, including retinoic acid, from the cytosol to the nucleus, where they activate nuclear receptors like peroxisome proliferator-activated receptor delta (PPARδ), thereby influencing cell proliferation and survival.
    Related references: PMID:21395585, PMID:22170058
  • By similarity, FABP5 modulates inflammation by regulating prostaglandin E2 (PGE2) biosynthesis via NF-kappa-B activation, suggesting a role in inflammatory responses.
  • May be involved in keratinocyte differentiation, consistent with its high expression in psoriatic skin.
    Related references: PMID:8092987
  • FABP5 is localized in the cytoplasm, nucleus, and at synapses (postsynaptic density); it is also secreted. In brain gray matter, it may serve as a synaptic carrier of endocannabinoids, controlling retrograde signaling.
    Related references: PMID:21395585
  • Post-translational modifications include acetylation, phosphorylation, and formation of disulfide bonds, which may regulate its lipid-binding and transport activities.

Experimental Guidance and Technical Tips

  • The immunogen spans the N-terminal region (aa 1–135), which is highly conserved across human, mouse, and rat, supporting the listed cross-reactivity. For novel sample types, validate cross-reactivity by sequence alignment or preliminary testing.
  • For Western blot, a band near 15 kDa is expected; use lysates from FABP5-expressing tissues such as skin or brain as positive controls.
  • For immunofluorescence (IF) and immunohistochemistry (IHC), consider using skin (preferably psoriatic) or brain sections; optimize fixation and permeabilization to detect secreted and intracellular pools.
  • For ELISA applications, confirm appropriate coating buffers and detection antibodies; consider pairing with a matched antibody to develop a sandwich ELISA for quantitative detection.
