Biological Background: FAAH2 Function and Localization
- Fatty-acid amide hydrolase 2 (FAAH2), also known as amidase domain-containing protein, anandamide amidohydrolase 2, or oleamide hydrolase 2, is encoded by the FAAH2 (AMDD) gene in humans.
- Catalyzes the hydrolysis of endogenous amidated lipids like the sleep-inducing lipid oleamide and the endocannabinoid anandamide, thereby regulating their signaling functions. Hydrolyzes monounsaturated anandamide preferentially over polyunsaturated substrates. Related references: PMID:17015445 PMID:19926788
- Subcellular localization includes membrane and lipid droplet, consistent with its role in lipid metabolism.
- Tissue expression is detected in kidney, liver, lung, prostate, heart, and ovary.
- Molecular weight is approximately 58 kDa; UniProt annotation lists a calculated mass of 58.3 kDa.
- Keywords: Hydrolase, Lipid degradation, Lipid droplet, Lipid metabolism, Transmembrane helix.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 323-532 of human FAAH2, a region that may aid in detecting full-length protein in samples where the enzyme is expressed.
- For western blotting, consider using whole cell lysates from tissues known to express FAAH2, such as kidney or liver, and validate the observed band size around 58 kDa.
- Immunofluorescence/immunocytochemistry may require optimization of fixation and permeabilization protocols due to the membrane and lipid droplet localization of FAAH2.
- ELISA experiments can be designed using purified or recombinant protein as a standard; however, confirm reactivity in your matrix of interest.
- Cross-reactivity with mouse and rat is reported; verify that the antibody performs comparably in these species under your experimental conditions.
