Biological Background: FAAH1 Function and Localization

  • Fatty-acid amide hydrolase 1 (FAAH1), also known as FAAH-1 or PSAB, is an enzyme responsible for the degradation of bioactive fatty acid amides.
  • Catalyzes the hydrolysis of endogenous amidated lipids, including the sleep-inducing lipid oleamide and the endocannabinoid anandamide, to their corresponding fatty acids, thereby regulating their signaling functions.
    Related references: PMID:17015445 PMID:19926788 PMID:9122178
  • Preferentially hydrolyzes the polyunsaturated substrate anandamide over monounsaturated substrates.
    Related references: PMID:17015445 PMID:9122178
  • Can also catalyze the hydrolysis of the endocannabinoid 2-arachidonoylglycerol.
    Related references: PMID:21049984
  • Cooperates with PM20D1 to hydrolyze N-fatty acyl amino acids, acting as a physiological regulator of specific subsets of intracellular N-fatty acyl amino acids.
  • Subcellularly localized to the endomembrane system, cytoplasm, and cytoskeleton.
  • Highly expressed in the brain, small intestine, pancreas, skeletal muscle, and testis; also detected in kidney, liver, lung, placenta, and prostate.
  • Contains a transmembrane domain and is subject to phosphorylation, classifying it as a phosphoprotein involved in lipid degradation.

Experimental Guidance and Technical Tips

  • The immunogen used for this antibody corresponds to amino acids 450-579 of human FAAH1. Researchers may consider using this antibody to detect the C-terminal region of FAAH1.
  • For Western blot applications, a band at approximately 63 kDa is expected. Validate the antibody in a relevant sample system, such as human or mouse brain lysate, where FAAH1 is highly expressed.
  • When performing ELISA, consider using recombinant FAAH1 protein as a positive control to assess binding specificity.
  • As this is a polyclonal antibody, potential batch-to-batch variability should be taken into account. It is recommended to optimize experimental conditions empirically.
