Biological Background: FAAH1 Function and Localization
- FAAH1 catalyzes the hydrolysis of bioactive fatty acid amides such as the sleep-inducing oleamide and the endocannabinoid anandamide, preferentially hydrolyzing polyunsaturated substrates, thus terminating their signaling. Related references: PMID:17015445, PMID:19926788, PMID:9122178
- It also hydrolyzes 2-arachidonoylglycerol (2-AG), another endocannabinoid. Related references: PMID:21049984
- In cooperation with PM20D1, FAAH hydrolyzes intracellular N-fatty acyl amino acids, acting as a physiological regulator of these molecules (By similarity).
- Subcellularly, FAAH1 is associated with the endomembrane system, cytoplasm, and cytoskeleton.
- It is highly expressed in the brain, small intestine, pancreas, skeletal muscle, and testis, with lower expression in kidney, liver, lung, placenta, and prostate.
- Phosphorylation and direct protein sequencing evidence indicate post-translational regulation; it localizes to membrane via a transmembrane helix.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 480–579 of human FAAH1; design experiments considering that the antibody may recognize epitopes within this C-terminal region.
- For WB, expect a band at approximately 63 kDa. Use appropriate loading controls and validate the specificity in your sample type.
- For IF/ICC, optimize fixation and permeabilization protocols for endomembrane/cytosolic targets; consider using suitable organelle markers.
- Cross-reactivity with mouse and rat has been reported; verify detection in the relevant model systems prior to routine use.
